			<content:encoded><![CDATA[<p><font color="#000000" face="Arial" id="role_document" size="2">Memorial Sloan-Kettering Cancer Center (MSKCC) is preparing for what it calls &ldquo;a breakthrough clinical trial&rdquo; that will test a new drug that uses a MEK inhibitor to target ETV1 in combination with imatinib on a small group of newly diagnosed GIST patients with advanced disease.</p>
<p>	We hope to begin recruitment in August, said Dr. William Tap, Section Chief of Sarcoma Oncology at MSKCC. The Phase II trial will be small, with only about 45 participants, and will only have one site at MSKCC in New York City, he said. That&rsquo;s because &ldquo;we just want to show that this approach works,&rdquo; before recruiting for a larger scale trial, Dr. Tap said. There will be a Phase I portion of the trial to define the Phase II doses of the combination regimen.</p>
<p>	The trial is unique because most new GIST clinical trials recruit patients that already have taken imatinib, sunitinib or other drugs and have already developed resistance to those drugs. This trial will try to show that treatment with a combination of imatinib and the MEK inhibitor from the start will be even more effective in combating GIST and will reduce the likelihood of secondary mutations that cause resistance.</p>
<p>	Novartis Pharmaceuticals is co-developing the MEK inhibitor, MEK162, with Array Biopharma. MEK162 has been through Phase I trials and recently completed a Phase II trial in NRAS-mutant melanoma.</p>
<p>	Dr. Tap said the research that opened the way to this new approach was led by his colleague at MSKCC, Dr. Ping Chi. She was able to show that ETV1, a transcription factor, was essential to the survival of GIST; while it was hard to target ETV1 because it didn&rsquo;t have molecule-binding pockets that could be blocked by drugs, she found a way around this problem by discovering that MEK inhibitors could do that job.</p>
<p>	&ldquo;MEK inhibitors alone doesn&rsquo;t seem to be great, but by doing combination therapy with imatinib we get a dramatic response,&rdquo; Dr. Chi said. ETV1 is a master regulator that is highly expressed in all GISTS.</p>
<p>	&ldquo;Ping&rsquo;s discovery is one of the major discoveries that we have seen in GIST research since the discovery of KIT,&rdquo; Dr. Tap said.</font></p>

She told her parents that she wanted friends and relatives to send money and she would donate whatever was received to charity. True to her word she has donated more than $400.00. The recipients included the Gist Cancer Research Fund,&nbsp; the Long Beach, New York animal shelter struggling to care for the surge in dogs and cats made homeless by hurricane Sandy, and a local animal shelter.</span><br />

<p style="margin-bottom: 0in"><span style="font-size:12px;"><span style="font-family: arial, helvetica, sans-serif;">The spirit of Peter Thomas led the way and along with Riette and Dylan Thomas (and Wayne Thomas in tow), the riders began and ended a triumphant journey along the rugged South Africa coast. </span></span></p>

<p style="margin-bottom: 0in"><span style="font-size:12px;"><span style="font-family: arial, helvetica, sans-serif;">It all started in Gordon&rsquo;s Bay and concluded in Cape Town. It was the toughest and most challenging of the three <i>Peter Thomas Memorial Rides For A Cure. </i>It stretched the riders physically, mentally and emotionally. They crossed fourteen mountain passes and endured gale force headwinds that were so strong that it almost blew the riders off their bikes.</span></span></p>
<p style="margin-bottom: 0in">&nbsp;</p>
<p style="margin-bottom: 0in"><span style="font-size:12px;"><span style="font-family: arial, helvetica, sans-serif;">While each cyclist came for a different reason on this trip, the camaraderie and banter amongst the group was certainly a unique experience for many. You do have to ask yourself the question, why some come back to do this grueling ride every three years? Through all the adversity, this fantastic group of GIST Cyclists left their jobs, families and comforts of home to cycle 1,000 miles in 10 days to raise money for the GIST<span id="more-2777"></span>Cancer cause and they never gave up. They are truly an inspiration and do what they do graciously and quietly without fanfare, egotism or arrogance. The world should take notice of how so many different people, from so many parts of the world, can form such a strong bond of brotherhood, respect and love. What a wonderful world it would be!</span></span><a href="/wp-content/uploads/final.jpg" rel="thumbnail"><img align="left" alt="" border="1" class="alignleft size-medium wp-image-2858" height="200" hspace="10" src="/wp-content/uploads/final-300x200.jpg" title="final" vspace="5" width="300" /></a></p>

We should all remember the pain they had to face or face on a daily basis and we all know someone who has had or has cancer and we are so blessed to share this experience with a group of healthy people.&rdquo;</i></span></span></p>

These tumors occur in the gastrointestinal tract and initially can be successfully treated with the targeted therapy drug Gleevec, but rapidly develop resistance to the treatment.</span></p>
<div style="color: rgb(78, 86, 90); font-family: Arial, Helvetica, sans-serif; line-height: 20px; ">The $120,000 award supports the research of Anette Duensing, M.D., assistant professor of pathology,&nbsp;<a href="http://pitt.edu/" style="color: rgb(119, 27, 97); text-decoration: none; cursor: pointer; outline: invert none medium; " target="_blank">University of Pittsburgh School of Medicine</a>. According to Dr. Duensing, GIST research in her laboratory aims to better understand the biology of<span id="more-2771"></span> GIST responses to Gleevec, as well as the mechanisms underlying drug resistance. These studies provide the framework for developing more effective treatment options for GIST patients.</div>

<div style="color: rgb(78, 86, 90); font-family: Arial, Helvetica, sans-serif; line-height: 20px; ">This year&rsquo;s award pushes the total amount of money Dr. Duensing has received from the GIST Cancer Research Fund to more than $720,000. &ldquo;This generous funding makes up a substantial part of my laboratory&rsquo;s budget and has allowed us to continue investigating why GIST patients develop resistance to Gleevec,&rdquo; said Dr. Duensing.</div>

Manipulating levels of this protein in GIST cells results in a faster and more effective destruction of the tumor cells.</div>

Dr. Duensing&rsquo;s laboratory is one of only five institutes nationally that the fund supports.</div>
